Intact mass measurements and determination of phosphorylation sites of proteins by mass spectrometry
Within the framework of the Mentor-Pupil Program of the MedinProt, our aim was the application of mass spectrometry for structural characterization of proteins and for the identification of phosphorylation sites. During our joint research work, the participant PhD student became familiar with the most important MS techniques needed for his PhD research, and obtained a broader overview about modern mass spectrometric methodologies used for protein/peptide analysis in general. Our new results are part of a publication (Gógl et al. FEBS Journal, 2018). In this project we analyzed the C-terminal phosphorylation sites of RSK1 which are autophosphorylated upon activation. Moreover, using synthetic phosphopeptides we identified characteristic fragment ions for future quantitative measurements.
