UV workstation for protein analysis and proteomics
The equipment is a standalone UV detector and computer-assisted data acquisition/controlling unit, which can be coupled to a chromatographic system, as well as to a mass spectrometer. The equipment includes an analog/digital interface for analog UV data collection and conversion. The workstation can be coupled to the Jasco HPLC pumps as well as to two electrospray ionization, ion trap tandem mass spectrometers, available in our laboratory. Any other type of HPLC or MS instruments can be coupled to the workstation. The purchased equipment consists of a Jasco, UV-2075 UV/VIS detector (monochromatic, 190-600 nm), as well as a ChromNAV chromatographic workstation, including a Jasco LCNetII interface
The UV detector workstation will be used for protein analysis, proteomics and method development. The proposed project specifically targets proteins, involved in Rheumatoid arthritis, which is an endemic, chronic inflammatory disease. We aim the detailed investigation of in vitro citrullination of proteins, as well as peptides derived from proteins with high diagnostic value, such as fibrin(ogen), vimentin and filaggrin. The purchased workstation can be used for microscale isolation and purification of citrullinated proteins with HPLC. Our aim is to study the enzymatic citrullination of synthetic peptides and isolated proteins in detail using HPLC-UV-ESI-MS analysis.
